Composition
Each tablet contains 500 mg pyrazinamide.
Properties
Pyrazinamide is highly effective as a bactericidal drug against tubercle bacilli. It exerts its effect in vivo in acidic loci in the body where tubercle bacilli are found, for instance, within phagocytic cells and in caseous lesions.
It is also active against bacilli resistant to other antituberculous drugs. Because of its unique activity against tubercle bacilli in an acid environment .
Pyrazinamide is now considered to be an important first-line agent in the intensive phase of short term antituberculous regimens. The product is not against atypical mycobacteria.
Pharmacokinetics
Pyrazinamide is readily absorbed from the gastrointestinal tract. Peak serum concentrations occur about 2 hours after oral administration.
The half-life has been reported to be about 9 to 10 hours. Pyrazinamide is widely distributed in body fluids and tissues and difuses into CSF.
It is metabolised primarily in the liver and about 70% of the dose appears in the urine within 24 hours mainly as metabolites and 4 to 14% as unchanged drug.
There are no major changes in the blood kinetics of pyrazinamide, isoniazid and rifampicin when the three drugs are administered separately or in a fixed combination over a 2 month period of continuous daily drug administration.
